What Is DSIP Peptide?

DSIP stands for delta sleep-inducing peptide. It is a nonapeptide, nine amino acids long, with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu. It was isolated in 1977 by Swiss researchers Schoenenberger and Monnier, who took blood from rabbits in which sleep had been induced by electrical stimulation of the brain, transferred it to other rabbits, and recorded more delta waves, the slow brain waves of deep sleep, in the recipients. The peptide responsible was named after that effect.

DSIP has since been found in the brain, in peripheral tissues and in blood plasma, in humans as well as animals. What has not been found, despite decades of work, is a dedicated DSIP receptor or a gene that clearly codes for it, which is part of what keeps the delta sleep inducing peptide interesting to researchers.

What Does DSIP Do? How DSIP Works in Research

DSIP is studied across several systems, and the research picture is more varied than its name suggests:

  • Sleep architecture. The original work, and much that followed, examined DSIP's effect on slow-wave sleep in animals. Small human studies in the 1980s produced mixed results, and the sleep effect remains debated.
  • Stress responses. Research reports that DSIP modulates stress-related hormone signalling and has been examined in animal models of stress.
  • Neuromodulation. Studies suggest DSIP interacts with several neurotransmitter systems rather than acting through one receptor, which may explain why its effects vary between models.
  • Antioxidant activity. Some papers examine protective effects against oxidative stress in cells and tissues.
